Hi Stevob Thank you for getting in touch with your question. The test actually works just as well if you lie it down. The reason that we stipulate that the test is run standing up is to minimise the possibility of spillage of the buffer solution. There is excess buffer solution so the test can lose about half of the buffer and still work correctly. So, we say that standing the test upright while it runs is good, but not critical to the test operation and outcome. The presence of the control line is a critical part of the test.
